對(dui)蝦(xia)(xia)種苗企業(ye)的(de)生(sheng)存主要是依靠蝦(xia)(xia)苗養殖(zhi)效果(guo)來決(jue)定的(de),蝦(xia)(xia)苗成活率(lv)的(de)高低直接(jie)影(ying)響著產業(ye)鏈的(de)上上下下。單胞(bao)藻(zao)對(dui)于(yu)蝦(xia)(xia)苗早期是最適合(he)(he)的(de)餌料,如(ru)(ru)果(guo)培(pei)育得(de)當(dang)(dang),不僅可(ke)以節(jie)約成本,減少育苗水體環境負荷(he),尤(you)其是對(dui)于(yu)蝦(xia)(xia)苗的(de)發育打下堅(jian)實的(de)基礎,提高成活率(lv)。結合(he)(he)個(ge)人(ren)的(de)育苗感受,投(tou)(tou)喂單胞(bao)藻(zao)與不投(tou)(tou)喂的(de)區別還是有的(de),比如(ru)(ru)苗的(de)變態(tai)時間縮(suo)短(duan),活力(li)個(ge)頭大小(xiao),水質均優于(yu)未投(tou)(tou)喂單胞(bao)藻(zao)的(de)苗,當(dang)(dang)然如(ru)(ru)果(guo)培(pei)育不得(de)當(dang)(dang),反作用(yong)也有,比如(ru)(ru)投(tou)(tou)喂有害菌(jun)超標的(de)藻(zao)液,藻(zao)類老化,投(tou)(tou)喂過量等。所以合(he)(he)理(li)(li)運用(yong),方可(ke)收到理(li)(li)想效果(guo)。限于(yu)個(ge)人(ren)水平(ping),描述方法的(de)錯(cuo)誤不足之處(chu),懇請(qing)大家(jia)指正。
一 、單胞藻純培養室操作描述
儀器使用
(1) 超凈工作(zuo)臺
使(shi)用前提前開機,打開紫外線燈和(he)風機開關(guan)(guan)20分鐘。超凈(jing)臺面上(shang)不(bu)存放不(bu)必要(yao)的(de)物品(pin),保持工作區內的(de)凈(jing)化氣流不(bu)受干(gan)擾,工作時盡量(liang)避(bi)免(mian)作明顯(xian)干(gan)擾氣流的(de)動作;超凈(jing)臺保持光滑水(shui)平,光滑便(bian)于(yu)用酒精消毒(du)布(bu)擦,水(shui)平是(shi)倒培(pei)(pei)養基時使(shi)培(pei)(pei)養皿(min)內平板(ban)厚度一致(zhi)。工作時超凈(jing)臺周(zhou)圍空(kong)氣流動要(yao)緩慢(man),雜菌盡量(liang)減(jian)少(shao),所以要(yao)定(ding)(ding)期清(qing)潔消毒(du)保種(zhong)室內,關(guan)(guan)閉門(men)窗,盡可能(neng)減(jian)少(shao)雜菌數量(liang);空(kong)氣過濾(lv)網定(ding)(ding)期清(qing)潔。
(2)真空泵和溶劑過濾(lv)器及過濾(lv)膜
此環節過濾(lv)只是為單(dan)胞藻純培養室(shi)(保種,提純,擴培試管級(ji))準備清潔無(wu)菌海(hai)水。其他級(ji)別的培養無(wu)需此環節。
過(guo)(guo)濾(lv)膜使用(yong)0.2微米孔(kong)徑,不(bu)(bu)可用(yong)手拿,用(yong)鑷子選取。真(zhen)(zhen)空(kong)(kong)泵(beng)與(yu)溶劑過(guo)(guo)濾(lv)器之間通過(guo)(guo)合適口徑的橡膠(jiao)管連接緊密,每(mei)次過(guo)(guo)濾(lv)要注意濾(lv)膜的過(guo)(guo)濾(lv)效果(guo),防止濾(lv)孔(kong)阻塞影響(xiang)過(guo)(guo)濾(lv)效果(guo)。濾(lv)瓶(ping)內的濾(lv)液(ye)不(bu)(bu)能沒(mei)過(guo)(guo)濾(lv)板出(chu)水口,否則濾(lv)液(ye)將被抽吸入真(zhen)(zhen)空(kong)(kong)泵(beng),導(dao)致(zhi)真(zhen)(zhen)空(kong)(kong)泵(beng)損壞(huai)。
應該使(shi)濾(lv)(lv)板內(nei)(nei)的營養(yang)液過濾(lv)(lv)盡后(無水滴)才能停泵。要定期注意(yi)真(zhen)空(kong)泵內(nei)(nei)油(you)量,注意(yi)加油(you)。
(3) 恒溫(wen)鼓風干(gan)燥箱
設(she)(she)定(ding)(ding)溫(wen)度時。先按控溫(wen)儀(yi)功能鍵(jian)SET進入(ru)溫(wen)度設(she)(she)定(ding)(ding)狀(zhuang)態,此(ci)時SP設(she)(she)定(ding)(ding)顯(xian)示一閃一閃,要按移(yi)動位(wei)鍵(jian)配合加(jia)鍵(jian)或減鍵(jian),設(she)(she)定(ding)(ding)時先從(cong)分位(wei)數(shu)字設(she)(she)定(ding)(ding),一直到(dao)百位(wei)數(shu),設(she)(she)定(ding)(ding)結(jie)束需按一下(xia)功能鍵(jian)SET,此(ci)時干燥箱進入(ru)升溫(wen)狀(zhuang)態,加(jia)熱指(zhi)示燈亮,當箱內溫(wen)度接近設(she)(she)定(ding)(ding)溫(wen)度時,加(jia)熱指(zhi)示燈閃動多次,控制(zhi)進入(ru)恒(heng)溫(wen)狀(zhuang)態。
使用注意事項(xiang):物品不能堆得太滿,更不能堵住出風口,以免影響溫度均勻上升。若是有牛皮紙和保鮮膜時,溫度不能超過180度。取出物品時,箱內外溫差不宜過高,防止玻璃器皿炸裂。橡膠制品不能放入。
(4)加濕(shi)器
配合生化培養箱使用。
水源是蒸餾水;不應容(rong)器內(nei)帶水移動加濕器;加水時(shi)應在容(rong)器上表示的滿水線為止。
應(ying)注意的事項:平時(shi)留意觀察水(shui)箱的存水(shui)量,及時(shi)予以(yi)補充,遠離(li)熱源(yuan),腐蝕(shi)物。
(5)蒸汽消(xiao)毒器(qi)
容器加(jia)水時,水位(wei)一定要超過(guo)電(dian)熱(re)管(guan),最好是到3個支撐點的(de)下緣,如果連續使(shi)(shi)用,應(ying)在每次(ci)消毒前補(bu)足蒸發水量,不(bu)然少(shao)水會燒壞電(dian)熱(re)管(guan)。蓋上(shang)的(de)放(fang)氣軟(ruan)管(guan)插(cha)入消毒桶圓槽內,對(dui)正蓋和主(zhu)(zhu)體(ti)的(de)螺(luo)栓槽,依次(ci)將相對(dui)方的(de)翼型螺(luo)母予以旋緊,使(shi)(shi)蓋與(yu)主(zhu)(zhu)體(ti)密(mi)合,加(jia)熱(re)開始時,應(ying)將放(fang)氣閥(fa)與(yu)安全閥(fa)的(de)小手柄(bing)扳(ban)至(zhi)垂(chui)直(zhi)放(fang)氣方位(wei),必須待有較(jiao)急蒸汽噴出,即將小手柄(bing)扳(ban)回水平“關(guan)閉”方位(wei)。
當到達所需壓力和溫度,計算消(xiao)毒時(shi)間,消(xiao)毒畢后,關閉電源,待(dai)壓力指針指向0時(shi),將放氣(qi)閥(fa)的小手柄扳至垂直放氣(qi)方位(wei),氣(qi)放完(wan)后,打開容器蓋。物品消(xiao)毒后要迅速干燥。
應注意滅菌液體(ti)(ti)時,不(bu)(bu)(bu)超(chao)過盛液體(ti)(ti)容(rong)器(qi)體(ti)(ti)積的(de)(de)3/4,瓶塞(sai)不(bu)(bu)(bu)可(ke)用(yong)軟木塞(sai)或未打孔的(de)(de)橡膠塞(sai)。滅菌器(qi)內冷空(kong)氣(qi)(qi)必(bi)須完全排(pai)除。因為空(kong)氣(qi)(qi)的(de)(de)膨脹壓(ya)(ya)(ya)(ya)大(da)于水蒸(zheng)氣(qi)(qi)的(de)(de)膨脹壓(ya)(ya)(ya)(ya),當水蒸(zheng)氣(qi)(qi)含(han)冷空(kong)氣(qi)(qi)時,壓(ya)(ya)(ya)(ya)力(li)(li)(li)(li)表(biao)所表(biao)示的(de)(de)壓(ya)(ya)(ya)(ya)力(li)(li)(li)(li)是水蒸(zheng)氣(qi)(qi)壓(ya)(ya)(ya)(ya)力(li)(li)(li)(li)和(he)(he)部分冷空(kong)氣(qi)(qi)壓(ya)(ya)(ya)(ya)力(li)(li)(li)(li)之和(he)(he),不(bu)(bu)(bu)是水蒸(zheng)氣(qi)(qi)的(de)(de)實際(ji)壓(ya)(ya)(ya)(ya)力(li)(li)(li)(li),而同一壓(ya)(ya)(ya)(ya)力(li)(li)(li)(li)下的(de)(de)實際(ji)溫(wen)度(du),含(han)空(kong)氣(qi)(qi)的(de)(de)蒸(zheng)汽低于飽和(he)(he)蒸(zheng)汽,這樣導致達不(bu)(bu)(bu)了消毒效果(guo)。絕不(bu)(bu)(bu)可(ke)堵塞(sai)安(an)全閥出氣(qi)(qi)孔,必(bi)須留空(kong)位保證其暢(chang)通放氣(qi)(qi),發現超(chao)溫(wen)超(chao)壓(ya)(ya)(ya)(ya),立(li)即斷電。
蒸餾器
先關閉放水(shui)(shui)(shui)閥,打開(kai)進(jin)水(shui)(shui)(shui)閥,待(dai)溢水(shui)(shui)(shui)皮管有水(shui)(shui)(shui)流出(chu),方可(ke)接通電(dian)源,此時(shi)可(ke)暫時(shi)關閉進(jin)水(shui)(shui)(shui)閥,待(dai)出(chu)水(shui)(shui)(shui)皮管有蒸餾(liu)水(shui)(shui)(shui)流出(chu),打開(kai)進(jin)水(shui)(shui)(shui)閥,制(zhi)備蒸餾(liu)水(shui)(shui)(shui)。定(ding)期清(qing)洗蒸餾(liu)器內壁(bi),除去水(shui)(shui)(shui)垢,不(bu)然(ran)影響蒸餾(liu)水(shui)(shui)(shui)質量和使(shi)用效果。切不(bu)可(ke)關閉水(shui)(shui)(shui)閥制(zhi)備蒸餾(liu)水(shui)(shui)(shui),否則燒(shao)壞(huai)電(dian)熱管。
(6)鼓(gu)風機
定期(qi)更換吸氣口的過濾棉,異常情況,請電工檢查。
(7)空調(diao)
保(bao)種室若有生長(chang)藻(zao)種,需維持恒定溫度(du),反之關閉(bi)。一級,二級空調視外界氣溫調整開(kai)關。
二 、衛生消毒
工具衛生消毒
1、濾(lv)(lv)水筒式(shi)過(guo)濾(lv)(lv)器。筒式(shi)過(guo)濾(lv)(lv)器的濾(lv)(lv)芯應用(yong)一(yi)次(ci)更(geng)換(huan)一(yi)次(ci),用(yong)過(guo)的濾(lv)(lv)芯浸泡在1%鹽酸溶液中消毒12h以上(shang)。然(ran)后用(yong)洗潔精(jing)清(qing)洗,再(zai)用(yong)足(zu)量清(qing)水沖(chong)洗干(gan)凈,晾干(gan)備用(yong)。(濾(lv)(lv)芯規格(ge)應小于(yu)等于(yu)0.5微米(mi))。
2、試管(guan)(guan)(規格10毫(hao)升(sheng)到20毫(hao)升(sheng)),錐形瓶(50毫(hao)升(sheng)到200毫(hao)升(sheng)),500ml錐形瓶,2.5L瓶廣口瓶,移液(ye)管(guan)(guan),移液(ye)槍管(guan)(guan)/盒(he),玻璃氣(qi)管(guan)(guan),膠(jiao)氣(qi)管(guan)(guan),氣(qi)石(shi),溶劑(ji)過濾器,培養(yang)皿(min)。用(yong)過后(hou)應當用(yong)清(qing)水清(qing)洗(xi)(xi)后(hou)浸泡(pao)于1% 鹽酸12h以上(溶劑(ji)過濾器除外(wai),浸泡(pao)時(shi)相近大小器皿(min)分類放不同的消毒(du)桶,規格相差大,易導(dao)致相互碰撞碎爛)然后(hou)用(yong)洗(xi)(xi)潔精(jing)清(qing)洗(xi)(xi)(用(yong)毛(mao)刷清(qing)洗(xi)(xi)試管(guan)(guan)底部時(shi),不可(ke)太用(yong)力(li),否則底部易破裂(lie)),再用(yong)大量清(qing)水沖洗(xi)(xi)干(gan)凈,風干(gan)或曬(shai)干(gan),高壓滅菌后(hou),放入恒溫干(gan)燥(zao)箱(膠(jiao)氣(qi)管(guan)(guan),氣(qi)石(shi)除外(wai))干(gan)燥(zao)備(bei)用(yong)。
3、聚乙烯藻袋用于(yu)二級(ji)藻類(lei)藻種掛袋培(pei)養或(huo)3級(ji)擴培(pei)(一般使用盛裝藻液12升左右)。先用清水沖洗(xi)(xi)干凈(jing)內外,浸(jin)泡(pao)于(yu)1% 鹽酸12h以上,再(zai)用大(da)量清水沖洗(xi)(xi)干凈(jing),風干或(huo)曬干,備用。由于(yu)藻袋近似密(mi)封性,對于(yu)防止微生物污染有很好的效果(guo)。
4、毛巾(jin)(jin)、保種室(shi),一級和(he)二級毛巾(jin)(jin)不可(ke)混用(yong),所用(yong)毛巾(jin)(jin)后都應消毒清洗晾干后備用(yong)。
5、錫箔紙:除非粘上油污或污物以及破(po)損,都應重復使用。
6、牛皮紙:定期拿到(dao)烘(hong)箱烘(hong)干。
7、定期更(geng)換空氣過濾器(qi)(用于1,2級(ji)培養(yang)的(de)進氣過濾)的(de)濾芯(xin),濾芯(xin)已(yi)油化(hua)或沖散的(de)棄去。
8、定期更換鼓風機進風口的過濾棉。
9、對新購入的(de)器(qi)皿應先除(chu)去包裝沾染的(de)污垢后,用(yong)洗(xi)(xi)潔(jie)精刷洗(xi)(xi),流水(shui)沖凈,再浸泡(pao)于1%的(de)鹽酸12h,使游離的(de)堿性物質除(chu)去,再用(yong)清水(shui)沖洗(xi)(xi)干凈,風干或曬干備用(yong)。
室內衛生消毒
1、保種室(shi)每天早上用清(qing)水擦洗實驗臺面,地板,墻壁,木架(jia)后,開紫(zi)外燈(deng)消毒20分鐘,此時關掉空調,關掉日光燈(deng),由于紫(zi)外燈(deng)照射會產生(sheng)一定量的臭氧,對人體有害,所以(yi)消毒關閉(bi)紫(zi)外燈(deng)后人應隔一段時間再(zai)進去(qu)。進保種室(shi)換鞋(xie)。
2、一級和二級室每天(tian)清潔,接(jie)種(zhong)前接(jie)種(zhong)后承(cheng)載物框架(jia)都(dou)應用消毒液消毒。
3、門口消(xiao)毒池定(ding)期更換消(xiao)毒藥水(shui)(高錳酸鉀)。
4、常用消毒劑濃度 1%鹽酸,次氯酸鈉20ppm(有效氯),碘液(1000ppm),酒(jiu)精70-75%,高錳(meng)酸鉀1mg/L。
三 、接種
藻種固體培養基制備
所需燒(shao)杯(bei)(bei),玻棒須清洗干凈,錐形瓶和培養(yang)皿須消毒清洗滅菌(jun)烘干。將(jiang)稱好的(de)純化瓊(qiong)脂粉加入燒(shao)杯(bei)(bei)再加入一(yi)定量的(de)海水(shui)(shui),加熱(必(bi)須不(bu)停攪拌,以免瓊(qiong)脂糊底燒(shao)焦)至完全融化,加熱后的(de)瓊(qiong)脂培養(yang)基應當透明(ming)無顯(xian)著沉淀,若明(ming)顯(xian),可采(cai)用清潔(jie)的(de)白色(se)薄絨布(bu)(bu)或中間(jian)夾有薄層(ceng)吸(xi)水(shui)(shui)棉的(de)雙(shuang)層(ceng)紗布(bu)(bu)鋪在漏斗上趁熱過(guo)濾,融化畢后精(jing)確加入營(ying)養(yang)鹽(維生素B溶液須等(deng)培養(yang)基高(gao)壓滅菌(jun)后冷(leng)卻至50攝(she)氏度以下再加)。
培(pei)(pei)養(yang)基(ji)(ji)采(cai)用121攝氏(shi)度高壓蒸汽滅(mie)菌(jun)20至30分(fen)鐘(zhong)即(ji)(ji)可(ke),培(pei)(pei)養(yang)基(ji)(ji)滅(mie)菌(jun)后應立即(ji)(ji)取出,冷卻至50攝氏(shi)度以(yi)下,即(ji)(ji)加維(wei)生(sheng)素B溶液。將盛裝培(pei)(pei)養(yang)基(ji)(ji)的錐形(xing)瓶(ping)和(he)培(pei)(pei)養(yang)皿放在(zai)超(chao)凈工作(zuo)臺上,點燃酒精燈(deng),右手托起錐形(xing)瓶(ping)底(di),將瓶(ping)口在(zai)酒精燈(deng)稍加燒,然后迅速將培(pei)(pei)養(yang)基(ji)(ji)倒入培(pei)(pei)養(yang)皿中,以(yi)鋪(pu)滿皿底(di)為止(厚薄適(shi)中)。鋪(pu)放好后,放置(zhi)20-30分(fen)鐘(zhong),待培(pei)(pei)養(yang)基(ji)(ji)凝(ning)固(gu)后,即(ji)(ji)開始操作(zuo)。
若是(shi)做斜(xie)面培養(yang)基,則加入維生素B溶液后,將盛(sheng)裝(zhuang)培養(yang)基的錐形瓶和試管以(yi)及一根厚約(yue)1厘米的小木板放在超凈工作(zuo)臺上。分(fen)裝(zhuang)約(yue)為試管高度的1/3-2/3(視情況操作(zuo)),分(fen)裝(zhuang)量(liang)應以(yi)能形成(cheng)2/3底層和1/3斜(xie)面量(liang)為好,擺置(zhi)成(cheng)適當斜(xie)面,待其自然凝固。培養(yang)基制備好后,如發(fa)現破(po)裂,水(shui)分(fen)浸(jin)入,色(se)澤異常等均應棄(qi)去.
待凝固后即(ji)進行操(cao)作,現一(yi)般采用平板(ban)劃線接(jie)種(zhong)(zhong)和涂布接(jie)種(zhong)(zhong)。
平板劃線接種
接種(zhong)(zhong)(zhong)(zhong)環(huan)(huan)在(zai)酒(jiu)精燈(deng)火(huo)(huo)(huo)焰上充(chong)分(fen)燒紅后(hou),接種(zhong)(zhong)(zhong)(zhong)柄一(yi)(yi)(yi)邊轉動(dong)(dong)一(yi)(yi)(yi)邊慢慢來回通(tong)(tong)過火(huo)(huo)(huo)焰3次(ci),待冷卻至室溫方可(ke)挑取(qu)藻種(zhong)(zhong)(zhong)(zhong)(為保證藻種(zhong)(zhong)(zhong)(zhong)不(bu)(bu)被燙死(si),挑種(zhong)(zhong)(zhong)(zhong)前可(ke)將(jiang)接種(zhong)(zhong)(zhong)(zhong)環(huan)(huan)接觸培(pei)(pei)(pei)養(yang)(yang)(yang)(yang)皿內壁或(huo)培(pei)(pei)(pei)養(yang)(yang)(yang)(yang)基(ji)),迅速(su)接種(zhong)(zhong)(zhong)(zhong)至新培(pei)(pei)(pei)養(yang)(yang)(yang)(yang)基(ji)(劃(hua)(hua)(hua)線可(ke)采(cai)用(yong)(yong)斜(xie)線法,方格法,曲線法,放射法等(deng))。左(zuo)手握(wo)住平板(ban)(ban),用(yong)(yong)大(da)拇(mu)指(zhi)和(he)食(shi)指(zhi)稍稍抬起(qi)皿蓋(gai),同(tong)時(shi)(shi)靠近火(huo)(huo)(huo)焰周圍,右手持接種(zhong)(zhong)(zhong)(zhong)環(huan)(huan)伸入皿內,在(zai)平板(ban)(ban)上一(yi)(yi)(yi)個區域作(zuo)(zuo)劃(hua)(hua)(hua)線(依方法不(bu)(bu)同(tong),動(dong)(dong)作(zuo)(zuo)不(bu)(bu)同(tong))。劃(hua)(hua)(hua)線時(shi)(shi)使接種(zhong)(zhong)(zhong)(zhong)環(huan)(huan)與平板(ban)(ban)表面成30-40度(du)角輕(qing)輕(qing)接觸,以(yi)腕力在(zai)表面作(zuo)(zuo)較快(kuai)的滑動(dong)(dong)(注意(yi)不(bu)(bu)要劃(hua)(hua)(hua)破平板(ban)(ban)表面或(huo)伸進培(pei)(pei)(pei)養(yang)(yang)(yang)(yang)基(ji)內)。接種(zhong)(zhong)(zhong)(zhong)環(huan)(huan)挑種(zhong)(zhong)(zhong)(zhong)后(hou)和(he)劃(hua)(hua)(hua)線完抽出時(shi)(shi)均不(bu)(bu)可(ke)碰(peng)及皿壁/口(kou),更不(bu)(bu)能(neng)碰(peng)其(qi)它物體(ti)和(he)臺面。接種(zhong)(zhong)(zhong)(zhong)完后(hou),將(jiang)接種(zhong)(zhong)(zhong)(zhong)環(huan)(huan)從柄部至環(huan)(huan)端逐漸通(tong)(tong)過酒(jiu)精燈(deng)火(huo)(huo)(huo)焰滅(mie)菌,不(bu)(bu)能(neng)直(zhi)接燒環(huan)(huan),以(yi)免(mian)殘留在(zai)接種(zhong)(zhong)(zhong)(zhong)環(huan)(huan)上的藻體(ti)或(huo)菌體(ti)爆濺污染(ran)空(kong)間。上述操作(zuo)(zuo)完成后(hou),即用(yong)(yong)無菌封口(kou)膜封培(pei)(pei)(pei)養(yang)(yang)(yang)(yang)皿口(kou)(膜使用(yong)(yong)前須(xu)用(yong)(yong)酒(jiu)精消毒,紫(zi)外燈(deng)消毒,風(feng)干(gan)),封口(kou)時(shi)(shi),用(yong)(yong)左(zuo)手大(da)拇(mu)指(zhi)摁(en)住膜一(yi)(yi)(yi)邊末(mo)端,右手大(da)拇(mu)指(zhi)和(he)食(shi)指(zhi)捏住另(ling)一(yi)(yi)(yi)邊末(mo)端,迅速(su)繞培(pei)(pei)(pei)養(yang)(yang)(yang)(yang)皿口(kou)繞一(yi)(yi)(yi)圈,封口(kou)。
涂(tu)布接種
所(suo)用(yong)種液(ye)必須經(jing)過(guo)弧菌檢(jian)測(ce)合(he)格后方可使用(yong)。涂(tu)布棒在火(huo)焰上(shang)慢慢來回(hui)過(guo)火(huo)焰3次,冷(leng)卻(不可驟冷(leng)),將種液(ye)用(yong)移(yi)液(ye)槍(qiang)從試管(guan)(guan)吸出(chu)注入(ru)平板上(shang)面(槍(qiang)管(guan)(guan)不可碰(peng)及試管(guan)(guan)外壁,更不能碰(peng)及其(qi)它物品或臺(tai)面),迅(xun)速用(yong)涂(tu)布棒在表面作來回(hui)左(zuo)右(you)的涂(tu)布,讓藻液(ye)分布均勻,畢(bi)后,封口同平板劃線(xian)。
試管液體培養基接種
所用(yong)種液(ye)必須經(jing)過弧菌檢(jian)測合格后方可使用(yong)。火(huo)焰滅(mie)菌前試(shi)(shi)(shi)(shi)管(guan)口外壁不(bu)(bu)能有水滴(di)(易導(dao)致(zhi)受熱(re)(re)不(bu)(bu)均而破裂),燒試(shi)(shi)(shi)(shi)管(guan)口時,試(shi)(shi)(shi)(shi)管(guan)傾斜45度(du)角,不(bu)(bu)斷移動試(shi)(shi)(shi)(shi)管(guan),使試(shi)(shi)(shi)(shi)管(guan)口受熱(re)(re)均勻,滅(mie)菌時間(jian)3秒(miao)左(zuo)右。滅(mie)菌后不(bu)(bu)能驟(zou)冷(leng),防止試(shi)(shi)(shi)(shi)管(guan)破裂。畢后,用(yong)盛(sheng)裝營(ying)養液(ye)150或200ml的錐形瓶(也要(yao)(yao)滅(mie)菌瓶口)向(xiang)試(shi)(shi)(shi)(shi)管(guan)中傾倒(dao)營(ying)養液(ye),每次(ci)只能拿一個試(shi)(shi)(shi)(shi)管(guan)(注:若幾(ji)個試(shi)(shi)(shi)(shi)管(guan)對(dui)比時,可以同時拿在手里,用(yong)大拇指和手掌(zhang)),倒(dao)好(hao)后,同樣試(shi)(shi)(shi)(shi)管(guan)和錐形瓶口滅(mie)菌,試(shi)(shi)(shi)(shi)管(guan)塞也要(yao)(yao)滅(mie)菌,塞試(shi)(shi)(shi)(shi)管(guan)塞時不(bu)(bu)要(yao)(yao)移動試(shi)(shi)(shi)(shi)管(guan)迎接塞,以免(mian)試(shi)(shi)(shi)(shi)管(guan)在移動中納入可能含菌空氣,火(huo)焰旁(pang)為無菌。
所(suo)有試(shi)(shi)管(guan)(guan)分裝好(hao)后,即接(jie)種。用(yong)移液(ye)槍(qiang)選取移液(ye)量(試(shi)(shi)管(guan)(guan)口滅菌),注入新培(pei)(pei)養(yang)(yang)基,槍(qiang)管(guan)(guan)不(bu)可碰(peng)及管(guan)(guan)外壁,更不(bu)能碰(peng)及其它物品(pin)或臺面,一支槍(qiang)管(guan)(guan)只(zhi)能和(he)同一種液(ye)使(shi)用(yong),選取另一試(shi)(shi)管(guan)(guan)種液(ye)時(shi)必(bi)須(xu)更換槍(qiang)頭。接(jie)種畢后,選取合(he)適光照溫度培(pei)(pei)養(yang)(yang)。由于試(shi)(shi)管(guan)(guan)培(pei)(pei)養(yang)(yang)是靜置培(pei)(pei)養(yang)(yang),而在靜置條件下,藻細胞會聚集在試(shi)(shi)管(guan)(guan)底部,且藻細胞周圍會形成透(tou)過養(yang)(yang)分的屏障,營養(yang)(yang)鹽的攝入受(shou)阻,光照吸收(shou)不(bu)均,為使(shi)營養(yang)(yang)鹽攝入正常,藻細胞受(shou)光均勻(yun),需要定時(shi)搖(yao)置試(shi)(shi)管(guan)(guan)(手先噴消毒酒精,手腕使(shi)勁搖(yao),不(bu)要擺(bai)臂,不(bu)能倒置搖(yao))
三角瓶,500ml瓶,2.5L瓶液體培養基接種
三(san)角(jiao)瓶接(jie)(jie)種需(xu)(xu)無(wu)菌操作(zuo)。500ml瓶,2.5L瓶液(ye)體培(pei)養基(ji)接(jie)(jie)種均在(zai)室內衛生條(tiao)件下操作(zuo)。3種瓶和所需(xu)(xu)營養液(ye)及錫紙,移液(ye)管(guan),洗(xi)耳球,氣(qi)管(guan),氣(qi)石均需(xu)(xu)經消毒清(qing)洗(xi),滅菌后使用。三(san)角(jiao)瓶需(xu)(xu)定時搖置(zhi),500ml,2.5L瓶培(pei)養在(zai)通氣(qi)條(tiao)件下培(pei)養。
二級聚乙烯藻袋液體培養基接種
氣管(guan)(guan),氣石均需經(jing)消毒清洗,滅(mie)菌后使(shi)用(yong)(yong)(yong)(yong),抽(chou)藻(zao)管(guan)(guan)前次用(yong)(yong)(yong)(yong)畢后應當用(yong)(yong)(yong)(yong)水泵(beng)將管(guan)(guan)內(nei)灌(guan)滿消毒鹽酸溶液(ye),今次用(yong)(yong)(yong)(yong)前需用(yong)(yong)(yong)(yong)海水或淡水沖洗干凈(jing)后使(shi)用(yong)(yong)(yong)(yong)。選取的(de)(de)種液(ye)應當是無明顯(xian)沉淀,泡沫(mo),原生動物(wu),附著物(wu),色澤正常(chang),藻(zao)細(xi)胞密(mi)度大,形態(tai)正常(chang),弧菌檢(jian)測合格(ge)后選用(yong)(yong)(yong)(yong)。所用(yong)(yong)(yong)(yong)海水漂白(bai)(bai)水消毒有效氯應足10ppm,曝氣12小時(shi)(shi)(shi),檢(jian)測余氯,加硫(liu)代硫(liu)酸鈉中和,方可(ke)使(shi)用(yong)(yong)(yong)(yong)。若使(shi)用(yong)(yong)(yong)(yong)稀釋(shi)的(de)(de)漂白(bai)(bai)水,應當天配制當天使(shi)用(yong)(yong)(yong)(yong),而(er)未使(shi)用(yong)(yong)(yong)(yong)的(de)(de)要(yao)在24小時(shi)(shi)(shi)后棄去。且稀釋(shi)的(de)(de)漂白(bai)(bai)水必(bi)須加蓋放在避光的(de)(de)容器內(nei)保(bao)存。由于(yu)漂白(bai)(bai)水會隨時(shi)(shi)(shi)間漸(jian)漸(jian)分(fen)解,一次性購入的(de)(de)漂白(bai)(bai)水不應太多(duo),不要(yao)過量儲存,以免影響滅(mie)菌功效。

1、母(mu)液(ye)的配置可以參(can)考(kao)教科書上所寫(xie),然后(hou)結合(he)當地的水(shui)質適當修改(gai)即可。氮,磷,鐵元(yuan)素(su),維(wei)(wei)生(sheng)素(su)B1,B12是必須使用,維(wei)(wei)生(sheng)素(su)母(mu)液(ye)保(bao)存于低(di)溫弱光環境下,其中(zhong)維(wei)(wei)生(sheng)素(su)的添加(jia)需(xu)要先高壓滅菌后(hou)再加(jia)入母(mu)液(ye)中(zhong)。其余營(ying)養(yang)鹽(yan)先配置好后(hou),再滅菌。如果是硅藻類,還應加(jia)上硅酸鈉。

一般情況下(xia)3袋二級藻(zao)(zao)種應先(xian)擴(kuo)培(pei)接種于容(rong)量400升的(de)桶(tong)(海水(shui)(shui)貯備及營養液提前配置好),然(ran)后立即(ji)進行二級藻(zao)(zao)袋擴(kuo)種,每個聚乙烯袋加(jia)18升左右藻(zao)(zao)水(shui)(shui)。
4、保種(zhong)培養基制備:按1.5%的比例加瓊脂粉,煮沸后加營養鹽(母液千分之1添加),高(gao)壓滅菌(jun)。維生素(su)需高(gao)壓滅菌(jun)后培養基冷卻至30攝(she)氏(shi)度方可加入(ru)。(可用劃(hua)線法(fa),涂布法(fa)等分離(li)培養藻種(zhong))。
5、儀(yi)器的(de)清洗(xi)維護(hu)。溶(rong)劑過濾器使(shi)用后(hou)只用洗(xi)潔精和清水清洗(xi)。高壓(ya)滅菌后(hou)備(bei)用。其它玻璃儀(yi)器,氣管,氣石,移液槍(qiang)筒(tong)使(shi)用后(hou)需要浸泡在百分(fen)之一到(dao)百分(fen)之十的(de)稀鹽酸溶(rong)液中至(zhi)少半天(tian),才能(neng)拿出(chu)清洗(xi),風干,高壓(ya)滅菌后(hou)備(bei)用。

三級藻生產
三(san)級藻(zao)系統(tong)包括用水(shui)處理以(yi)及配送,溫度與光照控制,藻(zao)種的(de)供(gong)應等。為確保(bao)擴大培(pei)養(yang)成功,從(cong)接種到收獲的(de)培(pei)養(yang)時(shi)間應該盡量縮短,以(yi)避(bi)免(mian)因為開(kai)放(fang)式培(pei)養(yang)隨培(pei)養(yang)規(gui)模擴大接觸外界(jie)感染源時(shi)間加長,三(san)級藻(zao)池滋生過多的(de)有害生物,使三(san)級培(pei)養(yang)失敗。
