蝦苗廠藻類三級培養技術,看看育苗得下多少工夫

對(dui)蝦(xia)(xia)(xia)種苗(miao)(miao)企業的(de)生存主(zhu)要(yao)是依靠(kao)蝦(xia)(xia)(xia)苗(miao)(miao)養殖效(xiao)果來決定的(de),蝦(xia)(xia)(xia)苗(miao)(miao)成活(huo)率(lv)的(de)高(gao)低直接(jie)影(ying)響著產業鏈的(de)上上下下。單(dan)胞(bao)藻(zao)對(dui)于蝦(xia)(xia)(xia)苗(miao)(miao)早期是最適合的(de)餌料,如(ru)果培(pei)育(yu)(yu)(yu)得(de)當,不(bu)僅可以節(jie)約(yue)成本,減少育(yu)(yu)(yu)苗(miao)(miao)水體(ti)環境負荷,尤其是對(dui)于蝦(xia)(xia)(xia)苗(miao)(miao)的(de)發育(yu)(yu)(yu)打下堅實的(de)基礎(chu),提高(gao)成活(huo)率(lv)。結合個人的(de)育(yu)(yu)(yu)苗(miao)(miao)感受,投喂(wei)單(dan)胞(bao)藻(zao)與不(bu)投喂(wei)的(de)區別(bie)還是有(you)的(de),比如(ru)苗(miao)(miao)的(de)變態(tai)時間縮短,活(huo)力(li)個頭大(da)小,水質(zhi)均(jun)優于未投喂(wei)單(dan)胞(bao)藻(zao)的(de)苗(miao)(miao),當然(ran)如(ru)果培(pei)育(yu)(yu)(yu)不(bu)得(de)當,反作用(yong)(yong)也有(you),比如(ru)投喂(wei)有(you)害菌超標的(de)藻(zao)液(ye),藻(zao)類老化,投喂(wei)過量(liang)等。所以合理運用(yong)(yong),方(fang)可收到理想效(xiao)果。限于個人水平,描述方(fang)法(fa)的(de)錯誤不(bu)足之處(chu),懇請(qing)大(da)家指正。

一 、單胞藻純培養室操作描述

儀器使用

(1) 超凈工作臺

使用前提前開機,打開紫外線(xian)燈(deng)和(he)風機開關20分(fen)鐘(zhong)。超凈臺面上不存放不必(bi)要(yao)的物品,保持(chi)工(gong)(gong)作(zuo)區(qu)內(nei)(nei)的凈化氣(qi)流(liu)(liu)不受干擾(rao),工(gong)(gong)作(zuo)時(shi)盡量避免作(zuo)明顯干擾(rao)氣(qi)流(liu)(liu)的動作(zuo);超凈臺保持(chi)光(guang)滑水平(ping),光(guang)滑便于(yu)用酒(jiu)精消(xiao)毒(du)布擦,水平(ping)是(shi)倒培養基時(shi)使培養皿內(nei)(nei)平(ping)板厚度一致(zhi)。工(gong)(gong)作(zuo)時(shi)超凈臺周圍空氣(qi)流(liu)(liu)動要(yao)緩慢,雜菌盡量減少,所以要(yao)定期(qi)清潔消(xiao)毒(du)保種(zhong)室(shi)內(nei)(nei),關閉門窗(chuang),盡可能(neng)減少雜菌數量;空氣(qi)過濾網定期(qi)清潔。

(2)真(zhen)空泵和溶劑過(guo)濾(lv)器(qi)及過(guo)濾(lv)膜

此環節過濾只(zhi)是為單(dan)胞(bao)藻純(chun)培(pei)養室(shi)(保種,提(ti)純(chun),擴(kuo)培(pei)試管(guan)級)準備清(qing)潔無(wu)菌(jun)海水。其他級別的培(pei)養無(wu)需此環節。

過(guo)(guo)(guo)濾膜使用(yong)0.2微(wei)米孔徑(jing),不可用(yong)手拿,用(yong)鑷子選(xuan)取(qu)。真空泵與溶劑過(guo)(guo)(guo)濾器之間(jian)通過(guo)(guo)(guo)合適口(kou)(kou)徑(jing)的(de)橡膠管連接緊密,每次(ci)過(guo)(guo)(guo)濾要(yao)注意(yi)濾膜的(de)過(guo)(guo)(guo)濾效(xiao)果,防止濾孔阻塞(sai)影響過(guo)(guo)(guo)濾效(xiao)果。濾瓶內的(de)濾液(ye)不能(neng)沒過(guo)(guo)(guo)濾板出水(shui)口(kou)(kou),否則濾液(ye)將被抽吸入真空泵,導致真空泵損壞。

應該使濾板內的營(ying)養液過(guo)濾盡后(無水滴)才能停泵(beng)。要定期注(zhu)意(yi)真(zhen)空泵(beng)內油量,注(zhu)意(yi)加(jia)油。

(3) 恒溫鼓(gu)風干燥箱

設定溫(wen)(wen)度(du)(du)時。先(xian)按(an)控溫(wen)(wen)儀功能鍵(jian)SET進入(ru)(ru)溫(wen)(wen)度(du)(du)設定狀態,此時SP設定顯示一閃(shan)(shan)一閃(shan)(shan),要(yao)按(an)移(yi)動位鍵(jian)配合加(jia)(jia)鍵(jian)或減鍵(jian),設定時先(xian)從分位數(shu)(shu)字設定,一直(zhi)到百位數(shu)(shu),設定結束需(xu)按(an)一下功能鍵(jian)SET,此時干燥箱進入(ru)(ru)升溫(wen)(wen)狀態,加(jia)(jia)熱(re)指(zhi)示燈亮,當箱內(nei)溫(wen)(wen)度(du)(du)接近設定溫(wen)(wen)度(du)(du)時,加(jia)(jia)熱(re)指(zhi)示燈閃(shan)(shan)動多次,控制進入(ru)(ru)恒(heng)溫(wen)(wen)狀態。

使用注意(yi)事項:物(wu)(wu)品(pin)不(bu)能(neng)堆得太(tai)滿,更不(bu)能(neng)堵住出風口(kou),以免影響溫(wen)度(du)均勻上升(sheng)。若是(shi)有牛皮(pi)紙和保(bao)鮮膜(mo)時,溫(wen)度(du)不(bu)能(neng)超過180度(du)。取出物(wu)(wu)品(pin)時,箱內(nei)外溫(wen)差不(bu)宜過高,防止玻璃(li)器(qi)皿(min)炸裂。橡(xiang)膠制品(pin)不(bu)能(neng)放入。

(4)加濕器

配合生化(hua)培養箱使用(yong)。

水(shui)源(yuan)是蒸餾水(shui);不(bu)應容器(qi)內帶水(shui)移動加濕器(qi);加水(shui)時應在容器(qi)上表示的滿水(shui)線為止。

應注(zhu)意的(de)事項(xiang):平時(shi)留意觀察水箱的(de)存水量,及時(shi)予以(yi)補充,遠離(li)熱源,腐蝕物。

(5)蒸(zheng)汽消毒器

容器加水時(shi),水位一定要超過(guo)電(dian)(dian)熱管,最好(hao)是到3個支撐點的(de)(de)下緣,如果連續使(shi)用,應在每(mei)次消毒(du)前補足(zu)蒸發水量,不然少水會燒壞電(dian)(dian)熱管。蓋(gai)上的(de)(de)放(fang)(fang)(fang)氣軟管插(cha)入(ru)消毒(du)桶圓槽內(nei),對(dui)正蓋(gai)和主體(ti)的(de)(de)螺栓槽,依(yi)次將(jiang)(jiang)相對(dui)方(fang)的(de)(de)翼型螺母(mu)予以旋緊(jin),使(shi)蓋(gai)與主體(ti)密合,加熱開(kai)始時(shi),應將(jiang)(jiang)放(fang)(fang)(fang)氣閥與安全閥的(de)(de)小(xiao)手(shou)柄(bing)扳至(zhi)垂(chui)直放(fang)(fang)(fang)氣方(fang)位,必須待有較(jiao)急蒸汽(qi)噴出,即將(jiang)(jiang)小(xiao)手(shou)柄(bing)扳回(hui)水平“關閉(bi)”方(fang)位。

當(dang)到達所需壓力(li)和溫度,計(ji)算消(xiao)毒時(shi)間,消(xiao)毒畢后(hou),關(guan)閉電源,待壓力(li)指針指向0時(shi),將放(fang)(fang)氣(qi)閥的小手柄(bing)扳至垂直放(fang)(fang)氣(qi)方(fang)位(wei),氣(qi)放(fang)(fang)完后(hou),打開容器蓋。物品消(xiao)毒后(hou)要迅(xun)速干燥。

應(ying)注意滅菌液體時,不(bu)超過盛液體容器體積的(de)(de)3/4,瓶塞(sai)(sai)(sai)不(bu)可(ke)(ke)用(yong)軟木(mu)塞(sai)(sai)(sai)或未(wei)打孔的(de)(de)橡膠塞(sai)(sai)(sai)。滅菌器內冷(leng)空(kong)氣(qi)必須(xu)完全(quan)(quan)排除。因為空(kong)氣(qi)的(de)(de)膨脹壓(ya)大于(yu)水蒸(zheng)(zheng)(zheng)氣(qi)的(de)(de)膨脹壓(ya),當水蒸(zheng)(zheng)(zheng)氣(qi)含(han)冷(leng)空(kong)氣(qi)時,壓(ya)力表所(suo)表示(shi)的(de)(de)壓(ya)力是水蒸(zheng)(zheng)(zheng)氣(qi)壓(ya)力和部分冷(leng)空(kong)氣(qi)壓(ya)力之和,不(bu)是水蒸(zheng)(zheng)(zheng)氣(qi)的(de)(de)實(shi)際(ji)壓(ya)力,而(er)同一(yi)壓(ya)力下的(de)(de)實(shi)際(ji)溫度,含(han)空(kong)氣(qi)的(de)(de)蒸(zheng)(zheng)(zheng)汽低于(yu)飽和蒸(zheng)(zheng)(zheng)汽,這樣導致達不(bu)了消(xiao)毒效(xiao)果(guo)。絕不(bu)可(ke)(ke)堵塞(sai)(sai)(sai)安(an)全(quan)(quan)閥出氣(qi)孔,必須(xu)留空(kong)位保(bao)證其暢通放(fang)氣(qi),發現超溫超壓(ya),立即斷電。

蒸餾器

先(xian)關閉放水(shui)(shui)(shui)閥,打開(kai)進(jin)水(shui)(shui)(shui)閥,待(dai)溢水(shui)(shui)(shui)皮(pi)(pi)管(guan)(guan)有水(shui)(shui)(shui)流出(chu),方可接通電源,此時可暫時關閉進(jin)水(shui)(shui)(shui)閥,待(dai)出(chu)水(shui)(shui)(shui)皮(pi)(pi)管(guan)(guan)有蒸(zheng)餾(liu)(liu)水(shui)(shui)(shui)流出(chu),打開(kai)進(jin)水(shui)(shui)(shui)閥,制備蒸(zheng)餾(liu)(liu)水(shui)(shui)(shui)。定期(qi)清洗蒸(zheng)餾(liu)(liu)器內壁,除(chu)去水(shui)(shui)(shui)垢,不(bu)然影(ying)響蒸(zheng)餾(liu)(liu)水(shui)(shui)(shui)質量和使用效果。切(qie)不(bu)可關閉水(shui)(shui)(shui)閥制備蒸(zheng)餾(liu)(liu)水(shui)(shui)(shui),否則燒壞電熱管(guan)(guan)。

(6)鼓風機

定(ding)期(qi)更換(huan)吸氣口的過濾棉,異常情況,請電工(gong)檢查。

(7)空(kong)調

保種室若(ruo)有生長藻(zao)種,需(xu)維持(chi)恒(heng)定溫度,反(fan)之關閉。一級,二級空調(diao)視外(wai)界氣(qi)溫調(diao)整(zheng)開關。

二 、衛生消毒

工具衛生消毒

1、濾(lv)水(shui)筒式過(guo)濾(lv)器。筒式過(guo)濾(lv)器的濾(lv)芯應用(yong)(yong)一次更(geng)換一次,用(yong)(yong)過(guo)的濾(lv)芯浸泡(pao)在1%鹽(yan)酸溶液(ye)中消毒12h以上。然后用(yong)(yong)洗潔精清(qing)洗,再(zai)用(yong)(yong)足量(liang)清(qing)水(shui)沖洗干凈,晾干備用(yong)(yong)。(濾(lv)芯規格(ge)應小于(yu)等(deng)于(yu)0.5微米(mi))。

2、試(shi)管(guan)(guan)(規格10毫(hao)升到20毫(hao)升),錐形瓶(50毫(hao)升到200毫(hao)升),500ml錐形瓶,2.5L瓶廣口(kou)瓶,移液管(guan)(guan),移液槍管(guan)(guan)/盒,玻(bo)璃(li)氣(qi)(qi)(qi)管(guan)(guan),膠(jiao)氣(qi)(qi)(qi)管(guan)(guan),氣(qi)(qi)(qi)石,溶劑過濾器(qi)(qi),培養皿(min)。用(yong)(yong)過后(hou)應當用(yong)(yong)清(qing)水清(qing)洗后(hou)浸泡(pao)于1% 鹽酸12h以上(溶劑過濾器(qi)(qi)除外(wai),浸泡(pao)時相(xiang)(xiang)近大小器(qi)(qi)皿(min)分類(lei)放(fang)不(bu)同的消毒桶,規格相(xiang)(xiang)差(cha)大,易導(dao)致相(xiang)(xiang)互碰(peng)撞碎爛)然后(hou)用(yong)(yong)洗潔(jie)精清(qing)洗(用(yong)(yong)毛刷清(qing)洗試(shi)管(guan)(guan)底(di)部時,不(bu)可(ke)太(tai)用(yong)(yong)力,否則(ze)底(di)部易破裂),再(zai)用(yong)(yong)大量清(qing)水沖洗干凈(jing),風干或(huo)曬干,高(gao)壓滅菌后(hou),放(fang)入恒溫干燥箱(膠(jiao)氣(qi)(qi)(qi)管(guan)(guan),氣(qi)(qi)(qi)石除外(wai))干燥備用(yong)(yong)。

3、聚(ju)乙烯藻(zao)(zao)袋用于(yu)(yu)二級藻(zao)(zao)類藻(zao)(zao)種(zhong)掛袋培養或3級擴培(一(yi)般使用盛裝(zhuang)藻(zao)(zao)液12升(sheng)左右(you))。先用清(qing)水沖(chong)洗干(gan)凈內外,浸泡于(yu)(yu)1% 鹽酸12h以上,再用大(da)量清(qing)水沖(chong)洗干(gan)凈,風干(gan)或曬干(gan),備用。由于(yu)(yu)藻(zao)(zao)袋近似密封性,對于(yu)(yu)防(fang)止微生物污染(ran)有很好的效果(guo)。

4、毛(mao)(mao)巾、保種室,一級(ji)(ji)和(he)二級(ji)(ji)毛(mao)(mao)巾不可混用(yong)(yong)(yong),所用(yong)(yong)(yong)毛(mao)(mao)巾后(hou)都應消毒清洗晾干后(hou)備(bei)用(yong)(yong)(yong)。

5、錫箔(bo)紙:除(chu)非粘上油污或污物以及破損,都應(ying)重復使用。

6、牛皮紙:定期拿到烘箱烘干。

7、定期更換空氣過濾器(qi)(用于1,2級(ji)培養(yang)的(de)(de)進氣過濾)的(de)(de)濾芯,濾芯已油化或(huo)沖散(san)的(de)(de)棄去。

8、定期更(geng)換鼓風機進風口(kou)的(de)過濾棉(mian)。

9、對新購入(ru)的器皿應先(xian)除去(qu)包(bao)裝沾染的污垢后,用(yong)洗潔(jie)精刷(shua)洗,流水(shui)沖凈,再浸泡于1%的鹽酸12h,使游離的堿性物質除去(qu),再用(yong)清水(shui)沖洗干凈,風(feng)干或曬(shai)干備用(yong)。

室內衛生消毒

1、保(bao)種(zhong)室每天早上(shang)用清水擦洗實驗臺面,地(di)板(ban),墻(qiang)壁(bi),木架后,開紫外(wai)燈消毒(du)20分鐘(zhong),此(ci)時關(guan)掉空調,關(guan)掉日光燈,由于紫外(wai)燈照射會產(chan)生(sheng)一定量的臭氧,對(dui)人體有害,所以消毒(du)關(guan)閉紫外(wai)燈后人應隔一段(duan)時間再(zai)進去(qu)。進保(bao)種(zhong)室換鞋。

2、一級和二級室每(mei)天清潔(jie),接種前接種后承載物框架都應用消毒(du)液消毒(du)。

3、門(men)口消毒池定期(qi)更換消毒藥水(高錳(meng)酸鉀(jia))。

4、常用消毒劑濃度(du) 1%鹽(yan)酸,次氯酸鈉20ppm(有效氯),碘液(ye)(1000ppm),酒精(jing)70-75%,高錳酸鉀1mg/L。

三 、接種

藻種固體培養基制備

所需燒(shao)(shao)(shao)杯,玻(bo)棒須(xu)清洗(xi)干凈(jing),錐形(xing)瓶和培養(yang)皿須(xu)消毒(du)清洗(xi)滅菌烘干。將稱好的(de)純化瓊(qiong)(qiong)(qiong)脂(zhi)粉(fen)加(jia)入燒(shao)(shao)(shao)杯再加(jia)入一(yi)定(ding)量的(de)海水,加(jia)熱(re)(必須(xu)不停攪拌,以免瓊(qiong)(qiong)(qiong)脂(zhi)糊底燒(shao)(shao)(shao)焦(jiao))至完全(quan)融化,加(jia)熱(re)后的(de)瓊(qiong)(qiong)(qiong)脂(zhi)培養(yang)基(ji)應當透明無顯著沉淀,若明顯,可采用(yong)清潔(jie)的(de)白色薄(bo)絨布或中間(jian)夾有(you)薄(bo)層吸水棉(mian)的(de)雙(shuang)層紗布鋪(pu)在(zai)漏斗上趁(chen)熱(re)過濾,融化畢(bi)后精確(que)加(jia)入營養(yang)鹽(維生素B溶液須(xu)等培養(yang)基(ji)高壓滅菌后冷卻至50攝氏(shi)度以下再加(jia))。

培(pei)養基(ji)采用(yong)121攝(she)氏度高壓蒸汽滅菌20至30分鐘(zhong)即可,培(pei)養基(ji)滅菌后應立即取(qu)出,冷卻至50攝(she)氏度以下,即加(jia)維生(sheng)素B溶(rong)液。將盛裝培(pei)養基(ji)的錐形(xing)瓶和培(pei)養皿(min)放在超凈(jing)工作臺上,點燃(ran)酒精(jing)燈,右手托起錐形(xing)瓶底,將瓶口在酒精(jing)燈稍(shao)加(jia)燒,然后迅速(su)將培(pei)養基(ji)倒(dao)入培(pei)養皿(min)中,以鋪滿(man)皿(min)底為止(厚(hou)薄適中)。鋪放好后,放置20-30分鐘(zhong),待培(pei)養基(ji)凝固后,即開始操作。

若是做斜面(mian)培養基,則加入(ru)維(wei)生素B溶液后,將盛裝培養基的(de)錐(zhui)形(xing)瓶和試管以及(ji)一根厚約(yue)1厘(li)米的(de)小木板放(fang)在超凈(jing)工作(zuo)臺上。分裝約(yue)為試管高度的(de)1/3-2/3(視情況操作(zuo)),分裝量應(ying)(ying)以能形(xing)成2/3底層和1/3斜面(mian)量為好(hao),擺(bai)置(zhi)成適當斜面(mian),待其自然凝固。培養基制(zhi)備好(hao)后,如發現破裂,水(shui)分浸入(ru),色澤異常等均應(ying)(ying)棄去.

待(dai)凝固后(hou)即進行操作,現一(yi)般采(cai)用平板劃(hua)線接種(zhong)和涂布接種(zhong)。

平板劃線接種

接(jie)(jie)種(zhong)(zhong)環(huan)在(zai)酒精燈(deng)火(huo)焰(yan)(yan)上充分燒紅后(hou),接(jie)(jie)種(zhong)(zhong)柄一邊(bian)(bian)轉動(dong)一邊(bian)(bian)慢慢來回通過(guo)火(huo)焰(yan)(yan)3次,待冷卻至室(shi)溫方(fang)可挑取藻(zao)種(zhong)(zhong)(為保證藻(zao)種(zhong)(zhong)不被燙(tang)死,挑種(zhong)(zhong)前(qian)可將(jiang)接(jie)(jie)種(zhong)(zhong)環(huan)接(jie)(jie)觸(chu)培(pei)(pei)養(yang)(yang)皿(min)(min)內(nei)壁(bi)或(huo)培(pei)(pei)養(yang)(yang)基(ji)),迅速接(jie)(jie)種(zhong)(zhong)至新(xin)培(pei)(pei)養(yang)(yang)基(ji)(劃(hua)(hua)線可采用(yong)斜線法(fa),方(fang)格法(fa),曲線法(fa),放(fang)射法(fa)等)。左手(shou)(shou)握住(zhu)平板,用(yong)大(da)拇指和食指稍稍抬起皿(min)(min)蓋,同(tong)時(shi)靠近火(huo)焰(yan)(yan)周圍,右(you)手(shou)(shou)持接(jie)(jie)種(zhong)(zhong)環(huan)伸入(ru)皿(min)(min)內(nei),在(zai)平板上一個(ge)區(qu)域作(zuo)(zuo)劃(hua)(hua)線(依方(fang)法(fa)不同(tong),動(dong)作(zuo)(zuo)不同(tong))。劃(hua)(hua)線時(shi)使(shi)(shi)接(jie)(jie)種(zhong)(zhong)環(huan)與平板表(biao)面(mian)(mian)成30-40度角輕輕接(jie)(jie)觸(chu),以腕力在(zai)表(biao)面(mian)(mian)作(zuo)(zuo)較(jiao)快的滑(hua)動(dong)(注意不要(yao)劃(hua)(hua)破平板表(biao)面(mian)(mian)或(huo)伸進培(pei)(pei)養(yang)(yang)基(ji)內(nei))。接(jie)(jie)種(zhong)(zhong)環(huan)挑種(zhong)(zhong)后(hou)和劃(hua)(hua)線完(wan)抽出時(shi)均不可碰及皿(min)(min)壁(bi)/口,更不能(neng)碰其它物體和臺面(mian)(mian)。接(jie)(jie)種(zhong)(zhong)完(wan)后(hou),將(jiang)接(jie)(jie)種(zhong)(zhong)環(huan)從(cong)柄部至環(huan)端逐(zhu)漸通過(guo)酒精燈(deng)火(huo)焰(yan)(yan)滅菌(jun),不能(neng)直接(jie)(jie)燒環(huan),以免殘留(liu)在(zai)接(jie)(jie)種(zhong)(zhong)環(huan)上的藻(zao)體或(huo)菌(jun)體爆濺污染空(kong)間(jian)。上述操作(zuo)(zuo)完(wan)成后(hou),即用(yong)無菌(jun)封(feng)口膜(mo)封(feng)培(pei)(pei)養(yang)(yang)皿(min)(min)口(膜(mo)使(shi)(shi)用(yong)前(qian)須用(yong)酒精消毒(du),紫外燈(deng)消毒(du),風干),封(feng)口時(shi),用(yong)左手(shou)(shou)大(da)拇指摁住(zhu)膜(mo)一邊(bian)(bian)末端,右(you)手(shou)(shou)大(da)拇指和食指捏(nie)住(zhu)另一邊(bian)(bian)末端,迅速繞培(pei)(pei)養(yang)(yang)皿(min)(min)口繞一圈,封(feng)口。

涂(tu)布(bu)接種(zhong)

所用種液(ye)必(bi)須經(jing)過弧菌檢測合格(ge)后方可使用。涂(tu)布(bu)棒(bang)在火焰(yan)上(shang)(shang)慢慢來回過火焰(yan)3次,冷(leng)卻(不可驟冷(leng)),將(jiang)種液(ye)用移液(ye)槍從試(shi)管(guan)吸出(chu)注入平板(ban)上(shang)(shang)面(槍管(guan)不可碰及(ji)試(shi)管(guan)外(wai)壁,更(geng)不能碰及(ji)其它物品(pin)或臺面),迅速用涂(tu)布(bu)棒(bang)在表(biao)面作來回左右的涂(tu)布(bu),讓藻液(ye)分布(bu)均勻(yun),畢后,封口(kou)同平板(ban)劃線。

試管液體培養基接種

所(suo)用種(zhong)液(ye)必(bi)須經(jing)過弧菌(jun)檢(jian)測合格后方可使(shi)用。火(huo)焰(yan)滅(mie)(mie)菌(jun)前試(shi)管(guan)(guan)口(kou)外壁(bi)不(bu)能(neng)有(you)水滴(di)(易導致受熱不(bu)均而破裂),燒試(shi)管(guan)(guan)口(kou)時(shi),試(shi)管(guan)(guan)傾斜45度角,不(bu)斷移動試(shi)管(guan)(guan),使(shi)試(shi)管(guan)(guan)口(kou)受熱均勻(yun),滅(mie)(mie)菌(jun)時(shi)間3秒左右。滅(mie)(mie)菌(jun)后不(bu)能(neng)驟冷,防止試(shi)管(guan)(guan)破裂。畢后,用盛裝營(ying)養(yang)液(ye)150或200ml的錐形(xing)瓶(也要(yao)滅(mie)(mie)菌(jun)瓶口(kou))向試(shi)管(guan)(guan)中傾倒營(ying)養(yang)液(ye),每次只能(neng)拿(na)一個試(shi)管(guan)(guan)(注:若幾(ji)個試(shi)管(guan)(guan)對(dui)比(bi)時(shi),可以同(tong)時(shi)拿(na)在手(shou)里,用大拇指(zhi)和手(shou)掌),倒好后,同(tong)樣試(shi)管(guan)(guan)和錐形(xing)瓶口(kou)滅(mie)(mie)菌(jun),試(shi)管(guan)(guan)塞(sai)也要(yao)滅(mie)(mie)菌(jun),塞(sai)試(shi)管(guan)(guan)塞(sai)時(shi)不(bu)要(yao)移動試(shi)管(guan)(guan)迎接塞(sai),以免試(shi)管(guan)(guan)在移動中納入可能(neng)含菌(jun)空氣,火(huo)焰(yan)旁為(wei)無菌(jun)。

所(suo)有試管(guan)(guan)分裝好后(hou),即(ji)接種。用移液(ye)槍選(xuan)取(qu)移液(ye)量(試管(guan)(guan)口滅菌),注入新培(pei)養(yang)基,槍管(guan)(guan)不(bu)(bu)(bu)可碰及(ji)管(guan)(guan)外(wai)壁,更不(bu)(bu)(bu)能碰及(ji)其它物品或臺(tai)面,一(yi)支槍管(guan)(guan)只能和(he)同一(yi)種液(ye)使(shi)用,選(xuan)取(qu)另一(yi)試管(guan)(guan)種液(ye)時必須更換槍頭。接種畢后(hou),選(xuan)取(qu)合適光(guang)照溫度培(pei)養(yang)。由于試管(guan)(guan)培(pei)養(yang)是靜置培(pei)養(yang),而(er)在靜置條件下,藻(zao)細胞(bao)會(hui)聚集在試管(guan)(guan)底部,且藻(zao)細胞(bao)周圍會(hui)形成透過養(yang)分的(de)屏障,營養(yang)鹽(yan)的(de)攝入受(shou)阻(zu),光(guang)照吸收不(bu)(bu)(bu)均,為使(shi)營養(yang)鹽(yan)攝入正常(chang),藻(zao)細胞(bao)受(shou)光(guang)均勻,需要定時搖(yao)置試管(guan)(guan)(手先噴(pen)消毒酒精,手腕使(shi)勁搖(yao),不(bu)(bu)(bu)要擺臂,不(bu)(bu)(bu)能倒置搖(yao))

三角瓶,500ml瓶,2.5L瓶液體培養基接種

三角瓶(ping)接種(zhong)需(xu)(xu)(xu)無菌操作。500ml瓶(ping),2.5L瓶(ping)液體(ti)培(pei)養(yang)基接種(zhong)均在室內衛生條(tiao)件下(xia)操作。3種(zhong)瓶(ping)和所需(xu)(xu)(xu)營(ying)養(yang)液及錫紙,移液管,洗(xi)耳球,氣管,氣石均需(xu)(xu)(xu)經(jing)消毒(du)清洗(xi),滅菌后使(shi)用。三角瓶(ping)需(xu)(xu)(xu)定時搖置,500ml,2.5L瓶(ping)培(pei)養(yang)在通氣條(tiao)件下(xia)培(pei)養(yang)。

二級聚乙烯藻袋液體培養基接種

氣(qi)(qi)管(guan),氣(qi)(qi)石均(jun)需經消(xiao)毒(du)清(qing)洗,滅菌后使(shi)用(yong),抽藻管(guan)前次用(yong)畢(bi)后應當用(yong)水(shui)泵(beng)將管(guan)內灌(guan)滿消(xiao)毒(du)鹽酸溶液,今次用(yong)前需用(yong)海(hai)水(shui)或(huo)淡水(shui)沖洗干凈后使(shi)用(yong)。選取的(de)(de)種液應當是(shi)無明顯沉(chen)淀,泡沫,原生(sheng)動物,附著物,色澤正常,藻細胞(bao)密(mi)度大,形態正常,弧(hu)菌檢(jian)測合格后選用(yong)。所(suo)用(yong)海(hai)水(shui)漂(piao)(piao)(piao)白水(shui)消(xiao)毒(du)有效氯(lv)應足10ppm,曝(pu)氣(qi)(qi)12小時(shi),檢(jian)測余氯(lv),加硫代硫酸鈉中和(he),方可使(shi)用(yong)。若(ruo)使(shi)用(yong)稀釋(shi)(shi)的(de)(de)漂(piao)(piao)(piao)白水(shui),應當天配制(zhi)當天使(shi)用(yong),而未使(shi)用(yong)的(de)(de)要(yao)在24小時(shi)后棄去(qu)。且稀釋(shi)(shi)的(de)(de)漂(piao)(piao)(piao)白水(shui)必須加蓋放(fang)在避光的(de)(de)容(rong)器內保存。由于漂(piao)(piao)(piao)白水(shui)會隨時(shi)間漸漸分(fen)解,一次性購入的(de)(de)漂(piao)(piao)(piao)白水(shui)不應太多,不要(yao)過量儲存,以免影響(xiang)滅菌功(gong)效。

1、母液(ye)(ye)的配(pei)置(zhi)可以(yi)參考教科書(shu)上(shang)所寫,然后(hou)結(jie)合當(dang)地的水(shui)質適當(dang)修改即可。氮,磷,鐵元(yuan)素,維生素B1,B12是(shi)必須使用,維生素母液(ye)(ye)保存于低溫弱光環境下(xia),其中維生素的添加需要先(xian)高壓滅菌(jun)(jun)后(hou)再(zai)加入(ru)母液(ye)(ye)中。其余營養鹽先(xian)配(pei)置(zhi)好后(hou),再(zai)滅菌(jun)(jun)。如果是(shi)硅藻類,還應加上(shang)硅酸鈉(na)。

三級(ji)藻(zao)系統包(bao)括用水處(chu)理以及配送(song),溫度與(yu)光(guang)照控(kong)制,藻(zao)種的(de)供應(ying)等。為(wei)確保擴大培養(yang)(yang)成(cheng)功,從接(jie)種到(dao)收獲的(de)培養(yang)(yang)時間應(ying)該盡量縮短,以避免因(yin)為(wei)開(kai)放(fang)式培養(yang)(yang)隨培養(yang)(yang)規模擴大接(jie)觸外界感染源時間加長,三級(ji)藻(zao)池滋生(sheng)過(guo)多的(de)有害生(sheng)物,使三級(ji)培養(yang)(yang)失敗。
