一 、單胞藻純培養室操作描述

儀器使用

(1) 超凈工(gong)作臺

使用前提前開機,打開紫(zi)外線燈和風機開關20分鐘。超(chao)(chao)(chao)凈(jing)臺面(mian)上(shang)不存放不必要的(de)物品,保(bao)持(chi)工作(zuo)(zuo)(zuo)(zuo)區內的(de)凈(jing)化氣流(liu)不受干擾,工作(zuo)(zuo)(zuo)(zuo)時(shi)盡(jin)量(liang)避(bi)免作(zuo)(zuo)(zuo)(zuo)明顯干擾氣流(liu)的(de)動作(zuo)(zuo)(zuo)(zuo);超(chao)(chao)(chao)凈(jing)臺保(bao)持(chi)光滑水(shui)平,光滑便于用酒精消毒(du)布擦,水(shui)平是倒培養(yang)基時(shi)使培養(yang)皿內平板厚度一致。工作(zuo)(zuo)(zuo)(zuo)時(shi)超(chao)(chao)(chao)凈(jing)臺周圍(wei)空(kong)氣流(liu)動要緩慢,雜菌盡(jin)量(liang)減少,所(suo)以要定(ding)期(qi)清潔消毒(du)保(bao)種室內,關閉門窗,盡(jin)可(ke)能減少雜菌數量(liang);空(kong)氣過(guo)濾(lv)網(wang)定(ding)期(qi)清潔。

(2)真空(kong)泵和溶劑(ji)過濾器及過濾膜

此環節過濾只是為單胞藻純培養(yang)室(保種(zhong),提純,擴培試管級(ji))準備(bei)清潔無(wu)菌海水。其(qi)他(ta)級(ji)別的培養(yang)無(wu)需此環節。

過(guo)濾(lv)(lv)膜(mo)使用(yong)0.2微米(mi)孔徑(jing),不可用(yong)手拿,用(yong)鑷子選(xuan)取。真空(kong)泵與溶劑(ji)過(guo)濾(lv)(lv)器之間(jian)通過(guo)合適(shi)口徑(jing)的橡膠管連接(jie)緊密,每次過(guo)濾(lv)(lv)要注意(yi)濾(lv)(lv)膜(mo)的過(guo)濾(lv)(lv)效果,防止濾(lv)(lv)孔阻塞影響過(guo)濾(lv)(lv)效果。濾(lv)(lv)瓶內的濾(lv)(lv)液不能(neng)沒過(guo)濾(lv)(lv)板出水口,否則濾(lv)(lv)液將被抽吸入(ru)真空(kong)泵,導致真空(kong)泵損壞。

應(ying)該使(shi)濾板內(nei)的營養液過濾盡后(無水滴)才(cai)能停泵(beng)(beng)。要定期注(zhu)意真空泵(beng)(beng)內(nei)油量,注(zhu)意加油。

(3) 恒溫鼓風干燥(zao)箱

設(she)(she)定(ding)溫(wen)(wen)度(du)時。先按(an)控溫(wen)(wen)儀功能(neng)鍵(jian)SET進入(ru)溫(wen)(wen)度(du)設(she)(she)定(ding)狀(zhuang)態,此(ci)時SP設(she)(she)定(ding)顯示(shi)(shi)一閃(shan)一閃(shan),要按(an)移動位(wei)鍵(jian)配合加(jia)鍵(jian)或減(jian)鍵(jian),設(she)(she)定(ding)時先從分位(wei)數字(zi)設(she)(she)定(ding),一直到百位(wei)數,設(she)(she)定(ding)結束(shu)需(xu)按(an)一下(xia)功能(neng)鍵(jian)SET,此(ci)時干(gan)燥箱進入(ru)升溫(wen)(wen)狀(zhuang)態,加(jia)熱指(zhi)示(shi)(shi)燈(deng)亮,當箱內(nei)溫(wen)(wen)度(du)接近設(she)(she)定(ding)溫(wen)(wen)度(du)時,加(jia)熱指(zhi)示(shi)(shi)燈(deng)閃(shan)動多次,控制(zhi)進入(ru)恒溫(wen)(wen)狀(zhuang)態。

使用(yong)注意事項:物(wu)品(pin)不(bu)能(neng)堆得太滿,更不(bu)能(neng)堵住(zhu)出風口,以免影響溫度(du)均勻上(shang)升(sheng)。若是有牛皮紙和保鮮膜時,溫度(du)不(bu)能(neng)超過(guo)180度(du)。取出物(wu)品(pin)時,箱內外(wai)溫差不(bu)宜過(guo)高,防止玻璃器皿炸裂。橡(xiang)膠制品(pin)不(bu)能(neng)放(fang)入。

(4)加濕器

配合生(sheng)化培(pei)養箱使用(yong)。

水(shui)源是蒸餾水(shui);不應(ying)容(rong)(rong)器(qi)內帶水(shui)移(yi)動加濕器(qi);加水(shui)時應(ying)在容(rong)(rong)器(qi)上表示(shi)的(de)滿水(shui)線為止。

應注意的事項:平時留意觀察水箱的存水量,及(ji)時予以補(bu)充,遠離熱(re)源,腐蝕物。

(5)蒸(zheng)汽消毒器(qi)

容器加水(shui)時,水(shui)位一定要超(chao)過電熱(re)管,最(zui)好是到3個(ge)支撐點的下緣,如(ru)果連續使用(yong),應(ying)在每次(ci)消(xiao)毒(du)前補足(zu)蒸發(fa)水(shui)量,不(bu)然(ran)少水(shui)會燒壞電熱(re)管。蓋(gai)上的放(fang)(fang)氣(qi)(qi)軟(ruan)管插(cha)入消(xiao)毒(du)桶圓(yuan)槽(cao)(cao)內,對正(zheng)蓋(gai)和主體的螺栓槽(cao)(cao),依次(ci)將相對方的翼型(xing)螺母予以旋緊,使蓋(gai)與主體密合,加熱(re)開始時,應(ying)將放(fang)(fang)氣(qi)(qi)閥與安(an)全閥的小手柄(bing)扳至垂(chui)直放(fang)(fang)氣(qi)(qi)方位,必須待有較急蒸汽噴出,即(ji)將小手柄(bing)扳回水(shui)平“關閉(bi)”方位。

當(dang)到達所需壓力(li)和溫度,計算消(xiao)毒時間(jian),消(xiao)毒畢后,關閉(bi)電源,待(dai)壓力(li)指(zhi)針指(zhi)向0時,將(jiang)放氣(qi)閥的小(xiao)手柄(bing)扳至(zhi)垂直放氣(qi)方位(wei),氣(qi)放完后,打開容器蓋。物品消(xiao)毒后要迅速(su)干燥。

應注意滅菌液體時,不(bu)超(chao)過盛液體容(rong)器體積(ji)的(de)3/4,瓶塞不(bu)可用(yong)軟木(mu)塞或未(wei)打孔的(de)橡膠塞。滅菌器內冷(leng)空(kong)氣(qi)(qi)(qi)(qi)必須(xu)完全(quan)排除。因為空(kong)氣(qi)(qi)(qi)(qi)的(de)膨脹壓(ya)(ya)(ya)大于水(shui)蒸(zheng)(zheng)(zheng)氣(qi)(qi)(qi)(qi)的(de)膨脹壓(ya)(ya)(ya),當水(shui)蒸(zheng)(zheng)(zheng)氣(qi)(qi)(qi)(qi)含冷(leng)空(kong)氣(qi)(qi)(qi)(qi)時,壓(ya)(ya)(ya)力(li)表所表示的(de)壓(ya)(ya)(ya)力(li)是水(shui)蒸(zheng)(zheng)(zheng)氣(qi)(qi)(qi)(qi)壓(ya)(ya)(ya)力(li)和(he)部(bu)分(fen)冷(leng)空(kong)氣(qi)(qi)(qi)(qi)壓(ya)(ya)(ya)力(li)之和(he),不(bu)是水(shui)蒸(zheng)(zheng)(zheng)氣(qi)(qi)(qi)(qi)的(de)實(shi)際壓(ya)(ya)(ya)力(li),而同一(yi)壓(ya)(ya)(ya)力(li)下的(de)實(shi)際溫度,含空(kong)氣(qi)(qi)(qi)(qi)的(de)蒸(zheng)(zheng)(zheng)汽低(di)于飽和(he)蒸(zheng)(zheng)(zheng)汽,這(zhe)樣導致達(da)不(bu)了消毒效果(guo)。絕不(bu)可堵塞安全(quan)閥(fa)出氣(qi)(qi)(qi)(qi)孔,必須(xu)留空(kong)位(wei)保證其(qi)暢通放氣(qi)(qi)(qi)(qi),發(fa)現超(chao)溫超(chao)壓(ya)(ya)(ya),立即斷電(dian)。

蒸餾器

先(xian)關閉放水(shui)(shui)閥(fa)(fa),打開(kai)(kai)進(jin)水(shui)(shui)閥(fa)(fa),待溢水(shui)(shui)皮(pi)管(guan)有水(shui)(shui)流出(chu)(chu),方可(ke)接通電(dian)源,此時(shi)(shi)可(ke)暫(zan)時(shi)(shi)關閉進(jin)水(shui)(shui)閥(fa)(fa),待出(chu)(chu)水(shui)(shui)皮(pi)管(guan)有蒸(zheng)餾(liu)水(shui)(shui)流出(chu)(chu),打開(kai)(kai)進(jin)水(shui)(shui)閥(fa)(fa),制(zhi)備蒸(zheng)餾(liu)水(shui)(shui)。定期清(qing)洗蒸(zheng)餾(liu)器(qi)內(nei)壁,除(chu)去(qu)水(shui)(shui)垢(gou),不(bu)然影響蒸(zheng)餾(liu)水(shui)(shui)質量和使用(yong)效果。切不(bu)可(ke)關閉水(shui)(shui)閥(fa)(fa)制(zhi)備蒸(zheng)餾(liu)水(shui)(shui),否則燒壞電(dian)熱(re)管(guan)。

(6)鼓風機

定(ding)期更換吸氣(qi)口的過濾棉,異常情況,請電工檢查。

(7)空調

保種(zhong)室若有生長(chang)藻種(zhong),需維持恒定(ding)溫度,反之關閉(bi)。一級(ji)(ji),二級(ji)(ji)空(kong)調視外界氣溫調整開關。

二 、衛生消毒

工具衛生消毒

1、濾(lv)(lv)水筒式(shi)過(guo)濾(lv)(lv)器(qi)。筒式(shi)過(guo)濾(lv)(lv)器(qi)的濾(lv)(lv)芯(xin)(xin)應用(yong)一次更換一次,用(yong)過(guo)的濾(lv)(lv)芯(xin)(xin)浸泡在1%鹽(yan)酸溶液中(zhong)消毒12h以上(shang)。然后(hou)用(yong)洗(xi)(xi)潔(jie)精(jing)清(qing)洗(xi)(xi),再用(yong)足量清(qing)水沖洗(xi)(xi)干(gan)凈(jing),晾干(gan)備用(yong)。(濾(lv)(lv)芯(xin)(xin)規格應小于(yu)等于(yu)0.5微米(mi))。

2、試管(guan)(guan)(規(gui)格(ge)10毫(hao)(hao)升到(dao)20毫(hao)(hao)升),錐(zhui)形瓶(ping)(50毫(hao)(hao)升到(dao)200毫(hao)(hao)升),500ml錐(zhui)形瓶(ping),2.5L瓶(ping)廣口瓶(ping),移液管(guan)(guan),移液槍(qiang)管(guan)(guan)/盒,玻璃氣(qi)(qi)管(guan)(guan),膠(jiao)氣(qi)(qi)管(guan)(guan),氣(qi)(qi)石,溶劑過濾(lv)器(qi),培養皿(min)。用(yong)(yong)過后(hou)應(ying)當用(yong)(yong)清(qing)(qing)水(shui)清(qing)(qing)洗(xi)后(hou)浸泡(pao)于1% 鹽酸12h以上(shang)(溶劑過濾(lv)器(qi)除(chu)外,浸泡(pao)時(shi)相(xiang)近大小器(qi)皿(min)分類放(fang)(fang)不(bu)同的(de)消(xiao)毒桶,規(gui)格(ge)相(xiang)差大,易導致(zhi)相(xiang)互碰(peng)撞碎(sui)爛)然后(hou)用(yong)(yong)洗(xi)潔(jie)精清(qing)(qing)洗(xi)(用(yong)(yong)毛刷(shua)清(qing)(qing)洗(xi)試管(guan)(guan)底(di)部(bu)時(shi),不(bu)可太用(yong)(yong)力(li),否則(ze)底(di)部(bu)易破裂),再用(yong)(yong)大量清(qing)(qing)水(shui)沖洗(xi)干(gan)凈,風干(gan)或曬干(gan),高壓滅菌后(hou),放(fang)(fang)入(ru)恒溫干(gan)燥箱(膠(jiao)氣(qi)(qi)管(guan)(guan),氣(qi)(qi)石除(chu)外)干(gan)燥備用(yong)(yong)。

3、聚乙烯藻(zao)袋(dai)用(yong)(yong)于(yu)二級(ji)(ji)藻(zao)類藻(zao)種掛(gua)袋(dai)培(pei)養或(huo)3級(ji)(ji)擴培(pei)(一(yi)般使用(yong)(yong)盛裝藻(zao)液(ye)12升左右)。先用(yong)(yong)清(qing)水沖洗干(gan)(gan)凈(jing)內外(wai),浸泡(pao)于(yu)1% 鹽酸12h以上(shang),再用(yong)(yong)大量清(qing)水沖洗干(gan)(gan)凈(jing),風干(gan)(gan)或(huo)曬干(gan)(gan),備用(yong)(yong)。由于(yu)藻(zao)袋(dai)近似(si)密封(feng)性,對(dui)于(yu)防止微生(sheng)物(wu)污染有很(hen)好的效果。

4、毛巾、保(bao)種(zhong)室(shi),一級和二級毛巾不(bu)可混用(yong),所用(yong)毛巾后都應消毒清洗(xi)晾(liang)干后備用(yong)。

5、錫箔紙:除(chu)非(fei)粘上油污(wu)或污(wu)物以及破損,都應重復使用。

6、牛皮紙:定期拿到烘箱(xiang)烘干。

7、定(ding)期(qi)更換空(kong)氣過(guo)濾器(用于1,2級培養的(de)進氣過(guo)濾)的(de)濾芯(xin),濾芯(xin)已油化或沖散的(de)棄去。

8、定期更換鼓風(feng)(feng)機進(jin)風(feng)(feng)口的過(guo)濾棉(mian)。

9、對新購入的器皿應先(xian)除去(qu)包裝沾染(ran)的污(wu)垢(gou)后,用洗潔精刷洗,流水沖(chong)凈,再浸泡(pao)于1%的鹽酸(suan)12h,使游離的堿性物質除去(qu),再用清(qing)水沖(chong)洗干凈,風干或曬干備(bei)用。

室內衛生消毒

1、保種室(shi)每天早上(shang)用清水擦洗實驗臺面(mian),地板,墻壁(bi),木(mu)架(jia)后,開(kai)紫(zi)(zi)外燈消毒20分鐘,此(ci)時(shi)(shi)關掉空調,關掉日光燈,由(you)于(yu)紫(zi)(zi)外燈照射會(hui)產生一(yi)(yi)定(ding)量(liang)的臭氧,對人(ren)體有害,所以(yi)消毒關閉紫(zi)(zi)外燈后人(ren)應隔一(yi)(yi)段時(shi)(shi)間再進去。進保種室(shi)換(huan)鞋(xie)。

2、一(yi)級(ji)和(he)二級(ji)室(shi)每(mei)天(tian)清潔,接種前接種后承載物框架都應(ying)用消毒(du)液消毒(du)。

3、門口(kou)消毒池定(ding)期(qi)更換消毒藥水(高錳酸鉀)。

4、常用消毒劑濃(nong)度(du) 1%鹽酸,次氯酸鈉20ppm(有效氯),碘(dian)液(1000ppm),酒精70-75%,高錳酸鉀1mg/L。

三 、接種

藻種固體培養基制備

所需燒杯(bei)(bei),玻棒須(xu)(xu)清洗干凈,錐形瓶和培養(yang)皿(min)須(xu)(xu)消毒清洗滅(mie)菌(jun)烘干。將稱好的(de)(de)純化(hua)瓊(qiong)(qiong)脂粉加入(ru)(ru)燒杯(bei)(bei)再(zai)加入(ru)(ru)一定量(liang)的(de)(de)海水,加熱(必須(xu)(xu)不停(ting)攪拌,以免瓊(qiong)(qiong)脂糊底燒焦)至(zhi)完(wan)全融(rong)(rong)化(hua),加熱后的(de)(de)瓊(qiong)(qiong)脂培養(yang)基應(ying)當透明無顯(xian)著沉淀,若(ruo)明顯(xian),可采(cai)用清潔的(de)(de)白(bai)色薄絨(rong)布或中間夾(jia)有薄層(ceng)吸水棉的(de)(de)雙層(ceng)紗布鋪在漏(lou)斗(dou)上趁熱過濾(lv),融(rong)(rong)化(hua)畢后精確(que)加入(ru)(ru)營(ying)養(yang)鹽(維(wei)生(sheng)素B溶液須(xu)(xu)等培養(yang)基高壓滅(mie)菌(jun)后冷卻至(zhi)50攝氏度以下再(zai)加)。

培(pei)(pei)養基采用121攝氏度(du)(du)高壓蒸汽滅(mie)菌20至30分(fen)鐘即可,培(pei)(pei)養基滅(mie)菌后應立即取出,冷卻至50攝氏度(du)(du)以下,即加維生(sheng)素B溶(rong)液(ye)。將(jiang)盛裝培(pei)(pei)養基的錐形瓶(ping)和培(pei)(pei)養皿(min)放在超凈工作臺上(shang),點燃(ran)酒精(jing)(jing)燈,右手托起錐形瓶(ping)底(di),將(jiang)瓶(ping)口(kou)在酒精(jing)(jing)燈稍(shao)加燒,然(ran)后迅(xun)速將(jiang)培(pei)(pei)養基倒入(ru)培(pei)(pei)養皿(min)中,以鋪(pu)滿皿(min)底(di)為止(zhi)(厚薄適(shi)中)。鋪(pu)放好后,放置20-30分(fen)鐘,待培(pei)(pei)養基凝固后,即開始操作。

若是做斜(xie)(xie)(xie)面(mian)培養基(ji),則加入(ru)維生(sheng)素(su)B溶液后(hou),將盛裝(zhuang)培養基(ji)的錐形(xing)瓶和試(shi)管以及一根(gen)厚約(yue)1厘米的小木(mu)板放在超凈工作臺上。分(fen)(fen)裝(zhuang)約(yue)為試(shi)管高度的1/3-2/3(視情況(kuang)操作),分(fen)(fen)裝(zhuang)量(liang)應以能形(xing)成2/3底層和1/3斜(xie)(xie)(xie)面(mian)量(liang)為好,擺置成適當斜(xie)(xie)(xie)面(mian),待其(qi)自(zi)然凝固(gu)。培養基(ji)制(zhi)備好后(hou),如發現破(po)裂,水分(fen)(fen)浸入(ru),色澤異常等均應棄去.

待凝(ning)固后即進行(xing)操作,現一般采(cai)用(yong)平板(ban)劃線接種和涂(tu)布接種。

涂布(bu)接(jie)種

所用(yong)種液(ye)必須經過(guo)(guo)弧菌檢測合(he)格后(hou)方可(ke)使用(yong)。涂布(bu)棒(bang)在火焰(yan)上慢慢來(lai)回過(guo)(guo)火焰(yan)3次,冷(leng)卻(不可(ke)驟冷(leng)),將種液(ye)用(yong)移(yi)液(ye)槍從試管(guan)(guan)吸出(chu)注入平(ping)板(ban)上面(mian)(槍管(guan)(guan)不可(ke)碰及(ji)試管(guan)(guan)外壁,更不能(neng)碰及(ji)其它物品或臺面(mian)),迅速用(yong)涂布(bu)棒(bang)在表面(mian)作來(lai)回左右(you)的涂布(bu),讓(rang)藻(zao)液(ye)分布(bu)均勻,畢(bi)后(hou),封(feng)口(kou)同平(ping)板(ban)劃(hua)線。

試管液體培養基接種

所用種液(ye)必須經過(guo)弧菌(jun)(jun)檢(jian)測合格后(hou)(hou)方可(ke)(ke)使用。火(huo)焰(yan)滅(mie)菌(jun)(jun)前試(shi)(shi)管(guan)(guan)(guan)口外壁不能(neng)(neng)有水滴(易導致受熱不均而(er)破裂),燒試(shi)(shi)管(guan)(guan)(guan)口時(shi),試(shi)(shi)管(guan)(guan)(guan)傾斜45度角(jiao),不斷移(yi)動試(shi)(shi)管(guan)(guan)(guan),使試(shi)(shi)管(guan)(guan)(guan)口受熱均勻,滅(mie)菌(jun)(jun)時(shi)間3秒左右。滅(mie)菌(jun)(jun)后(hou)(hou)不能(neng)(neng)驟冷,防止試(shi)(shi)管(guan)(guan)(guan)破裂。畢后(hou)(hou),用盛(sheng)裝營養(yang)液(ye)150或(huo)200ml的錐形瓶(ping)(也(ye)要(yao)(yao)滅(mie)菌(jun)(jun)瓶(ping)口)向試(shi)(shi)管(guan)(guan)(guan)中傾倒(dao)(dao)營養(yang)液(ye),每次只能(neng)(neng)拿一個試(shi)(shi)管(guan)(guan)(guan)(注(zhu):若(ruo)幾個試(shi)(shi)管(guan)(guan)(guan)對比時(shi),可(ke)(ke)以(yi)同(tong)時(shi)拿在手里,用大拇指和(he)手掌),倒(dao)(dao)好(hao)后(hou)(hou),同(tong)樣試(shi)(shi)管(guan)(guan)(guan)和(he)錐形瓶(ping)口滅(mie)菌(jun)(jun),試(shi)(shi)管(guan)(guan)(guan)塞(sai)也(ye)要(yao)(yao)滅(mie)菌(jun)(jun),塞(sai)試(shi)(shi)管(guan)(guan)(guan)塞(sai)時(shi)不要(yao)(yao)移(yi)動試(shi)(shi)管(guan)(guan)(guan)迎接塞(sai),以(yi)免試(shi)(shi)管(guan)(guan)(guan)在移(yi)動中納入可(ke)(ke)能(neng)(neng)含(han)菌(jun)(jun)空氣(qi),火(huo)焰(yan)旁為無(wu)菌(jun)(jun)。

所有試(shi)(shi)管(guan)分裝好后(hou),即接(jie)(jie)種(zhong)。用移(yi)液槍(qiang)選(xuan)(xuan)取移(yi)液量(liang)(試(shi)(shi)管(guan)口(kou)滅菌(jun)),注入新培(pei)(pei)養(yang)(yang)基(ji),槍(qiang)管(guan)不可碰(peng)及管(guan)外壁,更不能碰(peng)及其它物品(pin)或臺面,一支(zhi)槍(qiang)管(guan)只能和同一種(zhong)液使(shi)(shi)用,選(xuan)(xuan)取另一試(shi)(shi)管(guan)種(zhong)液時必須更換(huan)槍(qiang)頭(tou)。接(jie)(jie)種(zhong)畢后(hou),選(xuan)(xuan)取合適光(guang)照(zhao)溫度培(pei)(pei)養(yang)(yang)。由于(yu)試(shi)(shi)管(guan)培(pei)(pei)養(yang)(yang)是(shi)靜(jing)置培(pei)(pei)養(yang)(yang),而在(zai)靜(jing)置條件下,藻(zao)細胞會聚集在(zai)試(shi)(shi)管(guan)底部,且藻(zao)細胞周圍會形成透過(guo)養(yang)(yang)分的(de)(de)屏障(zhang),營養(yang)(yang)鹽的(de)(de)攝(she)入受阻,光(guang)照(zhao)吸收不均(jun),為使(shi)(shi)營養(yang)(yang)鹽攝(she)入正常(chang),藻(zao)細胞受光(guang)均(jun)勻,需要定(ding)時搖置試(shi)(shi)管(guan)(手先噴消毒酒精,手腕使(shi)(shi)勁搖,不要擺臂,不能倒置搖)

三角瓶,500ml瓶,2.5L瓶液體培養基接種

三角(jiao)瓶(ping)接(jie)種(zhong)需(xu)(xu)無菌(jun)操作。500ml瓶(ping),2.5L瓶(ping)液體(ti)培養基接(jie)種(zhong)均在室內衛(wei)生(sheng)條(tiao)件下操作。3種(zhong)瓶(ping)和所需(xu)(xu)營養液及(ji)錫紙,移液管,洗耳球,氣(qi)管,氣(qi)石(shi)均需(xu)(xu)經消毒清洗,滅菌(jun)后使用。三角(jiao)瓶(ping)需(xu)(xu)定時搖置,500ml,2.5L瓶(ping)培養在通氣(qi)條(tiao)件下培養。

二級聚乙烯藻袋液體培養基接種

氣管(guan),氣石均需經消毒(du)清(qing)洗,滅菌后使(shi)(shi)(shi)(shi)用(yong)(yong),抽藻(zao)管(guan)前次(ci)用(yong)(yong)畢后應(ying)(ying)(ying)(ying)當用(yong)(yong)水(shui)泵將(jiang)管(guan)內灌滿消毒(du)鹽(yan)酸溶液,今次(ci)用(yong)(yong)前需用(yong)(yong)海(hai)水(shui)或淡水(shui)沖洗干凈后使(shi)(shi)(shi)(shi)用(yong)(yong)。選取的種液應(ying)(ying)(ying)(ying)當是無明顯沉淀,泡(pao)沫,原生動物(wu),附著物(wu),色澤正(zheng)常(chang),藻(zao)細(xi)胞密度大(da),形態正(zheng)常(chang),弧菌檢測合格(ge)后選用(yong)(yong)。所用(yong)(yong)海(hai)水(shui)漂(piao)白(bai)(bai)水(shui)消毒(du)有(you)效氯(lv)應(ying)(ying)(ying)(ying)足10ppm,曝氣12小時,檢測余氯(lv),加(jia)硫代硫酸鈉中和(he),方可使(shi)(shi)(shi)(shi)用(yong)(yong)。若使(shi)(shi)(shi)(shi)用(yong)(yong)稀釋的漂(piao)白(bai)(bai)水(shui),應(ying)(ying)(ying)(ying)當天(tian)配制(zhi)當天(tian)使(shi)(shi)(shi)(shi)用(yong)(yong),而未(wei)使(shi)(shi)(shi)(shi)用(yong)(yong)的要(yao)在(zai)24小時后棄去(qu)。且稀釋的漂(piao)白(bai)(bai)水(shui)必須(xu)加(jia)蓋(gai)放在(zai)避(bi)光的容器(qi)內保存。由于(yu)漂(piao)白(bai)(bai)水(shui)會隨時間漸漸分解,一次(ci)性購(gou)入(ru)的漂(piao)白(bai)(bai)水(shui)不應(ying)(ying)(ying)(ying)太(tai)多,不要(yao)過量(liang)儲存,以免(mian)影(ying)響滅菌功(gong)效。

三級(ji)藻系統包(bao)括(kuo)用水處理以及配送,溫度與光照控(kong)制,藻種的供應等。為(wei)確保擴大培養(yang)(yang)成功,從(cong)接種到收獲(huo)的培養(yang)(yang)時(shi)間應該盡量縮短,以避免因為(wei)開(kai)放式(shi)培養(yang)(yang)隨(sui)培養(yang)(yang)規模擴大接觸外(wai)界感染源時(shi)間加長(chang),三級(ji)藻池滋(zi)生過(guo)多的有害生物,使三級(ji)培養(yang)(yang)失敗。
